    Near-infrared spectroscopy study of tourniquet-induced forearm ischaemia in patients with coronary artery disease

    Near-Infrared Spectroscopy (NIRS) can be employed to monitor local changes in haemodynamics and oxygenation of human tissues. A preliminary study has been performed in order to evaluate the NIRS transmittance response to induced forearm ischaemia in patients with coronary artery disease (CAD). The population consists in 40 patients with cardiovascular risk factors and angiographically documented CAD, compared to a group of 13 normal subjects. By inflating and subsequently deflating a cuff placed around the patient arm, an ischaemia has been induced and released, and the patients have been observed until recovery of the basal conditions. A custom NIRS spectrometer (IRIS) has been used to collect the backscattered light intensities from the patient forearm throughout the ischaemic and the recovery phase. The time dependence of the near-infrared transmittance on the control group is consistent with the available literature. On the contrary, the magnitude and dynamics of the NIRS signal on the CAD patients show deviations from the documented normal behavior, which can be tentatively attributed to abnormal vessel stiffness. These preliminary results, while validating the performance of the IRIS spectrometer, are strongly conducive towards the applicability of the NIRS technique to ischaemia analysis and to endothelial dysfunction characterization in CAD patients with cardiovascular risk factors

    A deep and wide-field view at the IC 2944/2948 complex in Centaurus

    We employed the ESO MPI wide-field camera and obtained deep images in the VIC passbands in the region of the IC 2944/2948 complex (l∼294.º 8; b∼−1.6º), and complemented them with literature and archival data. We used this material to derive the photometric, spectroscopic and kinematic properties of the brightest (V < 16) stars in the region. The VI deep photometry on the other end, helped us to unravel the lower main sequence of a few, possibly physical, star groups in the area. Our analysis confirmed previous suggestions that the extinction toward this line of sight follows the normal law (RV=3.1). We could recognize B-type stars spread in distance from a few hundred pc to at least 2 kpc. We found two young groups (age∼3 Myr) located respectively at about 2.3 and 3.2 kpc from the Sun. They are characterized by a significant variable extinction (E(B−V) ranging from 0.28 to 0.45 mag), and host a significant pre-main sequence population. We computed the initial mass functions for these groups and obtained slopes Gamma from -0.94 to -1.02 (e_Gamma=0.3), in a scale where the classical Salpeter law is -1.35. We estimated the total mass of both main stellar groups in ∼1100 and ∼500 M_sun, respectively. Our kinematic analysis indicated that both groups of stars deviate from the standard rotation curve of the Milky Way, in line with literature results for this specific Galactic direction. Finally, along the same line of sight we identified a third group of early-type stars located at ∼8 kpc from the Sun.     Neurodevelopmental disorders and development of project of life in a lifespan perspective: between habilitation and quality of life

    AbstractFor some years, the term “project of life” has become widely used in the field of neurodevelopmentaldisorders, and, at the same time, it has begun to make its way in many socialand health planning documents. However, beyond its relatively widespread use, this termdoes not yet possess an adequate and shared frame of the main underlying decision-makingprocesses. In particular, there is a need to identify the crucial questions for orientingthe choice of goals within the adolescent transition, which represents the complex hingebetween childhood and adulthood. Moreover, adulthood, which is often completely devoidof culturally and socially shared references, is still critical precisely because of thelack of future direction prompts usually represented by the stages of development. In thiscase, the themes of quality of life functioning as a guiding compass appear pertinent andmuch more relevant. The present contribution is, therefore, an attempt to present, in aunitary manner, the decision-making processes and questions at the basis of a constructof “project of life” shared within the scientific and associative communities

    The receptor-binding sequence of urokinase. A biological function for the growth-factor module of proteases.

    Previous studies have shown that the region of human urokinase-type plasminogen activator (uPA) responsible for receptor binding resides in the amino-terminal fragment (ATF, residues 1-135) (Stoppelli, M.P., Corti, A., Soffientini, A., Cassani, G., Blasi, F., and Assoian, R.K. (1985) Proc. Natl. Acad. Sci. U.S. A. 82, 4939-4943). The area within ATF responsible for specific receptor binding has now been identified by the ability of different synthetic peptides corresponding to different regions of the amino terminus of uPA to inhibit receptor binding of 125I-labeled ATF. A peptide corresponding to human [Ala19]uPA-(12-32) resulted in 50% inhibition of ATF binding at 100 nM. Peptides uPA-(18-32) and [Ala13]uPA-(9-20) inhibit at 100 and 2000 microM, respectively. The human peptide uPA-(1-14) and the mouse peptide [Ala20]uPA-(13-33) have no effect on ATF receptor binding. This region of uPA is referred to as the growth factor module since it shares partial amino acid sequence homology (residues 14-33) to epidermal growth factor (EGF). Furthermore, this region of EGF is responsible for binding of EGF to its receptor (Komoriya, A. Hortsch, M., Meyers, C., Smith, M., Kanety, H., and Schlessinger, J. (1984) Proc. Natl. Acad. Sci. U.S.A. 81, 1351-1355). However, EGF does not inhibit ATF receptor binding. Comparison of the sequences responsible for receptor binding of uPA and EGF indicate that the region of highest homology is between residues 13-19 and 14-20 of human uPA and EGF, respectively. In addition, there is a conservation of the spacings of four cysteines in this module whereas there is no homology between residues 20-30 and 21-33 of uPA and EGF. Thus, residues 20-30 of uPA apparently confer receptor binding specificity, and residues 13-19 provide the proper conformation to the adjacent binding region

    Detection and characterization of discontinuous motion on Thompson Glacier, Canadian High Arctic, using synthetic aperture radar speckle tracking and ice-flow modeling

    We investigate unusual discontinuous glacier motion on Thompson Glacier, Umingmat Nunaat, Arctic Canada, using synthetic aperture radar (SAR) images and ice-flow modeling. A novel intensity-rescaling scheme is developed to reduce errors in high-resolution speckle tracking, resulting in a ~25% improvement in accuracy. Interferometric SAR (InSAR) and speckle tracking using high resolution RADARSAT-2 data indicate velocity discontinuities of up to 1 cm d−1 across deep and longitudinally extensive supraglacial channels on Thompson Glacier. We use a cross-sectional finite-element ice-flow model to determine the conditions under which velocity discontinuities of the observed magnitude and signature are possible. The modeling suggests that discontinuous motion across (long and straight) supraglacial channels can occur without ice fracture and under a wide variety of glacier thermal structures, including in fully temperate glaciers. Despite the wide range of conditions conducive to discontinuous motion, the form we observe requires that the associated channels be deep, longitudinally extensive and located in regions of lateral shearing. We speculate that these combined conditions are rare except on polythermal glaciers, where drainage features such as moulins are comparatively scarce and lower deformation rates allow channels to incise consistently and persist over many years

    Rectal bleeding by Dieulafoy-like lesion: successful endoscopic treatment

    A 81-year old woman affected by chronic renal failure, non insulin-dependent diabetes mellitus (NIDM) and hypertension, had an severe anemia massive hematochezia. The colonoscopy could not localize the bleeding site except some blood spots in the rectum. The patient was readmitted after 1 month with hypovolemic shock by massive hematochezia and required several blood transfusions. The endoscopic examination showed an important arterial bleeding treated successfully with epinephrine and bipolar elettro-coagulation (BICAP). We suggested that the patient presented a Dieulafoy-like lesion; this is an uncommon gastrointestinal cause of bleeding due to a defect of a submucosal artery without evidence of atherosclerosis or vasculitis. Both chronic renal failure and age could be considered as predisponent factors in this patient. Hematochezia is the most important sign and is often complicated by haemorrhagic shock. The diagnosis was delayed due to the difficulty in localizing the bleeding site; moreover, the patient needed several blood transfusions. The arteriographic diagnosis associated to endoscopic treatment by epinephrine and BICAP enabled a successful therapy

    Comparative genomics and transcriptional profiles of Saccharopolyspora erythraea NRRL 2338 and a classically improved erythromycin over-producing strain

    <p>Abstract</p> <p>Background</p> <p>The molecular mechanisms altered by the traditional mutation and screening approach during the improvement of antibiotic-producing microorganisms are still poorly understood although this information is essential to design rational strategies for industrial strain improvement. In this study, we applied comparative genomics to identify all genetic changes occurring during the development of an erythromycin overproducer obtained using the traditional mutate-and- screen method.</p> <p>Results</p> <p>Compared with the parental <it>Saccharopolyspora erythraea </it>NRRL 2338, the genome of the overproducing strain presents 117 deletion, 78 insertion and 12 transposition sites, with 71 insertion/deletion sites mapping within coding sequences (CDSs) and generating frame-shift mutations. Single nucleotide variations are present in 144 CDSs. Overall, the genomic variations affect 227 proteins of the overproducing strain and a considerable number of mutations alter genes of key enzymes in the central carbon and nitrogen metabolism and in the biosynthesis of secondary metabolites, resulting in the redirection of common precursors toward erythromycin biosynthesis. Interestingly, several mutations inactivate genes coding for proteins that play fundamental roles in basic transcription and translation machineries including the transcription anti-termination factor NusB and the transcription elongation factor Efp. These mutations, along with those affecting genes coding for pleiotropic or pathway-specific regulators, affect global expression profile as demonstrated by a comparative analysis of the parental and overproducer expression profiles. Genomic data, finally, suggest that the mutate-and-screen process might have been accelerated by mutations in DNA repair genes.</p> <p>Conclusions</p> <p>This study helps to clarify the mechanisms underlying antibiotic overproduction providing valuable information about new possible molecular targets for rationale strain improvement.</p

    Protein fingerprints of cultured CA3-CA1 hippocampal neurons: comparative analysis of the distribution of synaptosomal and cytosolic proteins

    <p>Abstract</p> <p>Background</p> <p>All studies aimed at understanding complex molecular changes occurring at synapses face the problem of how a complete view of the synaptic proteome and of its changes can be efficiently met. This is highly desirable when synaptic plasticity processes are analyzed since the structure and the biochemistry of neurons and synapses get completely reshaped. Because most molecular studies of synapses are nowadays mainly or at least in part based on protein extracts from neuronal cultures, this is not a feasible option: these simplified versions of the brain tissue on one hand provide an homogeneous pure population of neurons but on the other yield only tiny amounts of proteins, many orders of magnitude smaller than conventional brain tissue. As a way to overcome this limitation and to find a simple way to screen for protein changes at cultured synapses, we have produced and characterized two dimensional electrophoresis (2DE) maps of the synaptic proteome of CA3-CA1 hippocampal neurons in culture.</p> <p>Results</p> <p>To obtain 2D maps, hippocampal cultures were mass produced and after synaptic maturation, proteins were extracted following subfractionation procedures and separated by 2D gel electrophoresis. Similar maps were obtained for the crude cytosol of cultured neurons and for synaptosomes purified from CA3-CA1 hippocampal tissue. To efficiently compare these different maps some clearly identifiable reference points were molecularly identified by mass spectrometry and immunolabeling methods. This information was used to run a differential analysis and establish homologies and dissimilarities in these 2D protein profiles.</p> <p>Conclusion</p> <p>Because reproducible fingerprints of cultured synapses were clearly obtained, we believe that our mapping effort could represent a simple tool to screen for protein expression and/or protein localization changes in CA3-CA1 hippocampal neurons following plasticity.</p
